Rats overexpressing the dopamine transporter display behavioral and neurobiological abnormalities with relevance to repetitive disorders

چکیده
The dopamine transporter (DAT) plays a pivotal role in maintaining optimal dopamine signaling. DAT-overactivity has been linked to various neuropsychiatric disorders yet so far the direct pathological consequences of it has not been fully assessed. We here generated a transgenic rat model that via pronuclear microinjection overexpresses the DAT gene. Our results demonstrate that DAT-overexpression induces multiple neurobiological effects that exceeded the expected alterations in the corticostriatal dopamine system. Furthermore, transgenic rats specifically exhibited behavioral and pharmaco-therapeutic profiles phenotypic of repetitive disorders. Together our findings suggest that the DAT rat model will constitute a valuable tool for further investigations into the pathological influence of DAT overexpression on neural systems relevant to neuropsychiatric disorders.

Brain serotonin and dopamine transporter bindings in adults with high-functioning autism.
CONTEXT Autism is a neurodevelopmental disorder that is characterized by repetitive and/or obsessive interests and behavior and by deficits in sociability and communication. Although its neurobiological underpinnings are postulated to lie in abnormalities of the serotoninergic and dopaminergic systems, the details remain unknown. OBJECTIVE To determine the occurrence of changes in the binding...

BACKGROUND A major risk-factor for developing Parkinson's disease (PD) is genetic variability in leucine-rich repeat kinase 2 (LRRK2), most notably the p.G2019S mutation. Examination of the effects of this mutation is necessary to determine the etiology of PD and to guide therapeutic development. OBJECTIVE Assess the behavioral consequences of LRRK2 p.G2019S overexpression in transgenic rats ...
متن کاملAssessing cocaine addiction vulnerability and neurobiological correlates within the nigrostriatal pathway using a novel model of dopamine D2 autoreceptor sensitivity
Cocaine addiction is a brain disorder that negatively affects the lives of millions of people and causes great societal harm. Most people who try cocaine do not become addicted to it. What predisposes someone to develop a cocaine dependency while others are able to control use is not well understood. This study’s aim is to use a novel rat model of dopamine D2 autoreceptor sensitivity to assess ...

Repeated periods of maternal separation in the early life of rats decreased dopamine transporter expression and significantly increased dopamine responses to stress, and behavioral responses to either stress or cocaine. As adults, maternal separation animals showed increased sensitivity to the effects of cocaine on locomotor activity and greater sensitivity to stress-induced sensitization to th...

Repetitive transcranial magnetic stimulation (rTMS) has been used widely in various psychiatric disorders like depression and schizophrenia. There have been some reports of its usefulness in alcohol dependence and substance use disorders. The present paper reviews the studies done using rTMS in substance use disorders including alcohol and nicotine dependence. Various studies done have been rev...
